Ligand-receptor interactions in chains of colloids:when reactions are limited by rotational diffusion

Zusammenfassung

We discuss (mostly theoretically) an experiment aimed at measuring the kinetics of biotin/streptavidin complex formation when both ligand and receptor are linked to the surface of colloids. Magnetic colloids (super paramagnetic) organize in chains in an external magnetic field, in the absence of binding reaction, colloids disassemble very fast after the field is turned off. Chains comprising a few mono-biotinated colloids dispersed among a majority of streptavinated colloids are formed during a time t. After dispersion of the chains, the fraction of colloids paired by the formation of a complex is measured by light scattering. The kinetics is found to be first order. The measured reaction time merely depends on the number of streptavidins per colloid and on the length of the spacer between the ligand (receptor) and the surface of the colloid. The results are rationalized in the frame of reaction-diffusion theory. We combine analytical results and more general simulation data.
