- (private-note)Canny 1997a, b, Huang etc. vessels were mostly frozen in the morning and in the evening, and they were mostly empty around noon. Daily cycles of cavitation and refilling. but... Cermak, Matyssek, Kucera 1993 found maximum sap flow to occur at noon. Minimum sapwood diameter reached at midday also leaves detached, put in plastic bags, taken to lab, leaflets detached, rachides either used or left standing in distilled water overnight to saturate. presssure chamber measurements of water potential on terminal leaflet (wrapped in alfoil and plastic bag before sampling) Freezing treatment: put in -25oC freezer, and left to cool. Thawing did not produce any signals. There were far more signals than conduits. The signals seemed to happen before the conduits could have been entirely frozen. This would mean several small bubbles might form in the one conduit. CryoSEM doesn't see the true filling state of conduits, because the water isn't vitrified; even the LN2 technique won't freeze water fast enough for that. in a cut xylem vessel air will fill to the cross wall
